PPP44mT is a novel thiosemicarbazone derivative being investigated for its anti-tumor activity, particularly in marginal zone lymphoma (MZL). It functions through a "double punch mechanism" that involves the high-affinity chelation of intracellular iron and copper, which are essential for tumor cell proliferation. This is followed by the formation of redox-active metal complexes that generate cytotoxic reactive oxygen species (ROS), leading to cell death. Preclinical studies have demonstrated that PPP44mT possesses potent activity (IC50 ~4.15 nM) and can overcome resistance to targeted agents such as BTK, PI3K, and BCL2 inhibitors. It was developed through collaboration between the Institute of Oncology Research in Switzerland and the Griffith Institute for Drug Discovery in Australia.
