Drug intelligence / Profile preview
PPRX-1701 is a nanoparticle formulation of 6-bromoindirubin acetoxime (BiA), a derivative of the traditional Chinese medicine indirubin. Developed by Phosphorex in collaboration with researchers at Brigham and Women's Hospital, it is designed to overcome the poor solubility of BiA for the treatment of glioblastoma. Mechanistically, BiA acts as a multi-kinase inhibitor targeting Src family kinases, glycogen synthase kinase-3 (GSK-3), and Janus kinase 2 (JAK2). Additionally, PPRX-1701 exerts immunotherapeutic effects by blocking the expression of immunosuppressive molecules such as indoleamine 2,3-dioxygenase 1 (IDO1) and PD-L1 downstream of interferon-gamma (IFNγ) signaling. In preclinical murine models of glioblastoma, intravenous administration of PPRX-1701 has demonstrated improved survival and favorable alterations in the tumor immune microenvironment, including reduced regulatory T cells and increased CD8+ T cells.
