Overview

PRAME mRNA refers to a therapeutic modality consisting of messenger RNA encoding the Preferentially Expressed Antigen in Melanoma (PRAME), a well-characterized cancer-testis antigen. As a therapeutic, PRAME mRNA is primarily developed as a component of mRNA-based cancer vaccines (such as BioNTech's BNT111) designed to stimulate an autologous immune response. Upon administration, the mRNA is taken up by cells, typically dendritic cells, and translated into the PRAME protein. This protein is then processed and presented on the cell surface via MHC molecules to activate PRAME-specific CD4+ and CD8+ T cells. This mechanism aims to eliminate PRAME-expressing tumor cells, which are prevalent in melanoma, uveal melanoma, acute myeloid leukemia, and various epithelial cancers. Beyond its therapeutic application, the measurement of endogenous PRAME mRNA levels serves as a critical diagnostic and prognostic biomarker, where high expression is often associated with advanced disease stages, increased metastatic potential, and poor clinical outcomes.
