Prednisone + ruxolitinib is a combination of two pharmaceutical agents used primarily in research and clinical settings for their synergistic effects, particularly in the treatment of myeloproliferative neoplasms such as myelofibrosis. Prednisone is a synthetic glucocorticoid corticosteroid with potent anti-inflammatory and immunosuppressive properties, acting mainly through glucocorticoid receptor agonism to modulate gene expression and suppress immune responses[5]. Ruxolitinib is an oral small molecule inhibitor that selectively targets Janus kinase 1 (JAK1) and Janus kinase 2 (JAK2), key mediators in the JAK/STAT signaling pathway involved in cytokine signaling, hematopoiesis, and inflammation[6][8]. The combination has been shown to inhibit both JAK/STAT (notably STAT5) and MAPK (ERK) pathways, resulting in reduced pro-inflammatory status and cell proliferation. This dual approach may enhance therapeutic efficacy against diseases characterized by aberrant immune activation or fibrosis[1][2][3].
