Prizloncabtagene autoleucel is an autologous, next-generation chimeric antigen receptor (CAR) T-cell therapy engineered to express a bispecific CAR targeting both the cluster of differentiation 19 (CD19) and cluster of differentiation 20 (CD20) antigens. These are tumor-associated antigens commonly expressed on B-cell malignancies. The product uses a second-generation CAR construct with a 4‑1BB costimulatory domain, designed to enhance T-cell activation and persistence. Its mechanism involves immunologic cytotoxicity against malignant B cells expressing either or both target antigens, leading to their elimination. Developed primarily for relapsed/refractory B-cell non-Hodgkin lymphoma (B-NHL), including large B-cell lymphoma and diffuse large B cell lymphoma, prizloncabtagene autoleucel has demonstrated high response rates and durable remissions in early-phase clinical trials[1][2][3][5].
