Overview

PT0511 is a small-molecule pan-KRAS degrader developed by PAQ Therapeutics for the treatment of KRAS-driven solid tumors, including lung, colorectal, and pancreatic cancers. The drug utilizes a proprietary Autophagy-Tethering Compound (ATTEC) platform, which represents a novel approach in targeted protein degradation (TPD). Unlike PROTACs that utilize the ubiquitin-proteasome system, PT0511 recruits the autophagy machinery by tethering the KRAS protein to LC3 (microtubule-associated protein 1 light chain 3), facilitating the sequestration of KRAS into autophagosomes and its subsequent degradation in lysosomes. As a pan-KRAS degrader, PT0511 is designed to target multiple mutant isoforms of KRAS as well as wild-type KRAS, potentially providing a broader therapeutic benefit and overcoming resistance mechanisms associated with allele-specific inhibitors.
