PTK7-CAR gamma delta T cells are an experimental cellular immunotherapy being developed for the treatment of T-cell acute lymphoblastic leukemia (T-ALL). This therapeutic approach utilizes gamma delta (γδ) T cells, which possess innate antitumor activity and MHC-independent antigen recognition, as a platform for chimeric antigen receptor (CAR) engineering. By targeting Protein Tyrosine Kinase 7 (PTK7)—a transmembrane pseudokinase overexpressed in T-ALL but minimally expressed in normal γδ T cells—this therapy aims to overcome common CAR-T challenges such as fratricide and T-cell aplasia. To enhance therapeutic efficacy, the γδ T cells are expanded ex vivo in the presence of transforming growth factor beta (TGFβ), which has been shown to improve their persistence and bone marrow trafficking in murine models.
