PTK7-targeted CAR-T cell therapy is an investigational cellular immunotherapy designed to target Protein Tyrosine Kinase 7 (PTK7), a pseudokinase receptor in the Wnt signaling pathway that is highly expressed on cancer stem cells (CSCs) and various solid tumors. The therapy involves engineering a patient's T cells to express a chimeric antigen receptor (CAR) that specifically recognizes PTK7, leading to antigen-specific activation and tumor cell lysis. Preclinical studies have demonstrated efficacy in models of triple-negative breast cancer (TNBC), non-small-cell lung cancer (NSCLC), ovarian cancer, and neuroblastoma, particularly those with ATRX in-frame fusions which drive high PTK7 expression.
