Overview

PTOC1 is an autologous, second-generation CD19-targeted chimeric antigen receptor T-cell (CAR-T) therapy developed by Chongqing Precision Biotech Co., Ltd. for the treatment of refractory systemic lupus erythematosus (SLE). The therapy involves the genetic modification of a patient's own T-cells to express a CAR that identifies and eliminates CD19-expressing B-cells, which are key drivers of the autoimmune response in SLE. PTOC1 utilizes an optimized preparation scheme designed to enhance efficacy and safety, potentially allowing for lower therapeutic doses and more durable clinical outcomes compared to earlier CAR-T generations. In December 2024, the therapy received clinical trial approval from China's NMPA for adult patients with refractory SLE.
