Overview

PU-H71 + ruxolitinib is an investigational combination therapy consisting of PU-H71, a selective inhibitor of the epichaperome component of the molecular chaperone heat shock protein 90 (HSP90), and ruxolitinib, an oral Janus kinase (JAK) 1 and 2 inhibitor. The combination targets two critical pathways in myelofibrosis and potentially other hematological malignancies: PU-H71 disrupts oncogenic protein networks stabilized by HSP90, while ruxolitinib blocks aberrant JAK-STAT signaling in malignant hematopoietic cells. PU-H71 is being developed by Samus Therapeutics. The primary indication under study is myelofibrosis, especially in patients with inadequate response to ruxolitinib monotherapy. The combination is undergoing early-phase clinical trials to evaluate safety, tolerability, and preliminary efficacy[1][3][5].
