Overview

PXS-5505 + ruxolitinib is a combination therapy consisting of PXS-5505, a first-in-class small molecule pan-lysyl oxidase inhibitor, and ruxolitinib, a Janus kinase (JAK) inhibitor. PXS-5505 targets the lysyl oxidase family of enzymes, which are critical in crosslinking collagen and contributing to fibrosis; ruxolitinib inhibits the JAK1 and JAK2 kinases, thus modulating inflammatory pathways and reducing symptom burden in myeloproliferative neoplasms. This combination is being investigated for the treatment of myelofibrosis, where PXS-5505 aims to address the fibrotic component of the disease while ruxolitinib addresses inflammatory and proliferative aspects. Preliminary clinical data have shown safety and potential efficacy in myelofibrosis, especially in patients with intermediate and high-risk disease who have been on stable ruxolitinib therapy[1][3].
