Drug intelligence / Profile preview
QTX3034 + cetuximab + oxaliplatin + leucovorin + fluorouracil is an investigational combination regimen consisting of the oral, G12D-preferring multi-KRAS inhibitor QTX3034, the anti-EGFR monoclonal antibody cetuximab, and the standard FOLFOX chemotherapy regimen (oxaliplatin, leucovorin, and fluorouracil). Developed by Quanta Therapeutics, this combination is designed to treat KRAS G12D-mutated cancers, particularly colorectal cancer (CRC). QTX3034 is a noncovalent, allosteric inhibitor that binds to the inactive (GDP-bound) state of KRAS, with preferential activity against the G12D mutant. The addition of cetuximab blocks compensatory EGFR feedback signaling that typically occurs upon KRAS inhibition, thereby enhancing the trapping of KRAS in its inactive state. The combination with FOLFOX chemotherapy provides additional cytotoxic anti-tumor activity. The regimen has received Fast Track designation from the FDA for the treatment of patients with KRAS G12D-mutated colorectal cancer and is being evaluated in a Phase 1 clinical trial.
