The ras peptide vaccine is an investigational cancer immunotherapy designed to stimulate the immune system to recognize and attack tumor cells harboring mutations in the RAS family of oncogenes, particularly KRAS. The most studied form is TG01, which consists of synthetic peptides corresponding to common mutant forms of KRAS. These peptides are administered with an adjuvant to induce a specific T-cell response against cancer cells expressing the mutated protein. The primary mechanism involves generating white blood cells that target and kill tumor cells with KRAS mutations. This approach has been explored mainly for pancreatic cancer and non-small cell lung cancer (NSCLC), where such mutations are prevalent[1][2][4]. Clinical trials have shown that the vaccine can induce immune responses in a significant proportion of patients and is generally well tolerated[2][3][5]. Development efforts have included both direct vaccination and ex vivo loading of antigen-presenting cells with mutant ras peptides[3]. While early-phase studies showed safety and immunogenicity, no significant survival benefit has yet been demonstrated in advanced disease settings; current research focuses on adjuvant use after standard therapy[2][4].
