Overview

rb79 is an experimental monoclonal antibody designed to target the TRPV6 (Transient Receptor Potential Vanilloid 6) calcium channel, which is frequently overexpressed in various cancers, particularly prostate cancer. It specifically binds to an extracellular epitope located on the loop between the S1 and S2 transmembrane segments of the channel. Unlike traditional antagonists, rb79 induces a complex biphasic response characterized by the transient activation of the TRPV6 channel, which subsequently potentiates store-operated calcium entry (SOCE). This disruption of intracellular calcium homeostasis leads to enhanced cell death via apoptosis. Preclinical studies have demonstrated that rb79 significantly decreases the survival of TRPV6-positive prostate cancer cell lines, such as LNCaP, while having no effect on TRPV6-deficient cells, suggesting its potential as a targeted oncological therapy.
