Overview

RG1503 is a synthetic glycosaminoglycan mimetic belonging to the class of ReGeneraTing Agents (RGTAs). It is engineered from a 40 kDa dextran backbone and designed to mimic the biological activity of heparan sulfate, a naturally occurring molecule involved in tissue repair and regeneration. The drug acts primarily by restoring extracellular matrix integrity and normalizing the balance between matrix metalloproteinases (MMPs) and their inhibitors (TIMPs), thereby promoting tissue healing. Preclinical studies have demonstrated that RG1503 can reverse degenerative changes in tissues affected by inflammation—such as those seen in periodontitis, Crohn’s disease, and chemotherapy-induced mucositis—by repairing collagen networks, reducing apoptosis in inflamed tissues, rebuilding bone structure, and protecting basement membranes. Its mechanism involves interaction with transforming growth factor β1 (TGF-β1), leading to anabolic effects on extracellular matrices[1][5][6].
