Overview

RG6505 is a small-molecule Pan-RAS inhibitor being developed by Roche, in collaboration with the University of Texas MD Anderson Cancer Center, for the treatment of advanced solid tumors. As a Pan-RAS inhibitor, RG6505 is designed to target and inhibit multiple isoforms and mutant versions of the RAS protein family, including KRAS, NRAS, and HRAS. This approach aims to overcome the limitations of allele-specific inhibitors (such as KRAS G12C inhibitors) by blocking signaling across a broader range of RAS-driven cancers and potentially addressing resistance mechanisms mediated by other RAS isoforms. The compound is currently undergoing evaluation in Phase 1 clinical trials to determine its safety, tolerability, and preliminary efficacy in patients with RAS-mutated malignancies.
