Overview

Ripertamab (SCT400) is a recombinant human-mouse chimeric anti-CD20 IgG1 monoclonal antibody developed by Sinocelltech. It targets the CD20 antigen on B lymphocytes, sharing the same antigen-binding sites and variable regions as rituximab but differing by a single amino acid at position 219 of the heavy chain constant region (valine instead of alanine). This modification incorporates the G1m(1,17) allotype, which is prevalent in Asian populations. Ripertamab induces B-cell depletion through antibody-dependent cell-mediated cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and direct apoptosis. It was approved in China in 2022 for the treatment of CD20-positive B-cell non-Hodgkin lymphoma, including diffuse large B-cell lymphoma (DLBCL). It is also being evaluated for autoimmune conditions such as primary membranous nephropathy (PMN) and immune thrombocytopenia (ITP).
