Overview

This is a multi-agent combination regimen consisting of the monoclonal antibody rituximab (anti-CD20), the alkylating agent cyclophosphamide, the vinca alkaloid vincristine (Oncovin), the corticosteroid prednisone, and an interferon (typically interferon-alpha). The regimen is used primarily in hematologic malignancies such as follicular lymphoma. Rituximab targets CD20 on B-cells leading to cell lysis; cyclophosphamide crosslinks DNA causing apoptosis; vincristine inhibits microtubule formation disrupting mitosis; prednisone induces apoptosis in lymphoid cells and has anti-inflammatory effects; and interferons have antiproliferative and immunomodulatory actions. This combination aims to maximize antitumor efficacy by targeting cancer cells through multiple mechanisms[2][3].
