This investigational combination therapy consists of the anti-CD20 monoclonal antibody rituximab (marketed as Rituxan or Mabthera) and fresh frozen plasma (FFP). Developed by Wolfson Medical Center, the regimen is specifically intended for the treatment of advanced refractory chronic lymphocytic leukemia (CLL). The therapeutic rationale is based on the observation that many CLL patients possess deficiencies in the complement system, which significantly limits the efficacy of rituximab by impairing complement-dependent cytotoxicity (CDC). By administering FFP as an exogenous source of complement proteins immediately prior to rituximab infusion, the protocol aims to restore and enhance the immune-mediated destruction of malignant B cells. Clinical evaluation in Phase II trials has focused on patients who are refractory to standard fludarabine-based treatments.
