The combination of rituximab and ibrutinib is an FDA-approved, chemotherapy-free regimen for the treatment of certain B-cell malignancies, including chronic lymphocytic leukemia (CLL), small lymphocytic lymphoma (SLL), and Waldenström's macroglobulinemia. Ibrutinib is a first-in-class, oral small-molecule inhibitor of Bruton's tyrosine kinase (BTK), which disrupts B-cell receptor signaling critical for malignant B-cell proliferation and survival. Rituximab is a monoclonal antibody targeting the CD20 antigen on B-cells, leading to cell lysis through immune-mediated mechanisms. The combination has demonstrated improved progression-free survival compared to standard regimens in clinical trials and offers a non-chemotherapy option for patients with these hematologic cancers[1][2][4][8].
