Overview

**rituximab + interferon alfa-2a** is a combination immunotherapy regimen consisting of the anti-CD20 monoclonal antibody rituximab and the cytokine interferon alfa-2a. Rituximab is a chimeric monoclonal antibody targeting the CD20 antigen on B-cells, leading to B-cell depletion through antibody-dependent cellular cytotoxicity (ADCC), complement-mediated cytotoxicity (CDC), phagocytosis, and direct signaling-induced apoptosis[2]. Interferon alfa-2a is a recombinant cytokine that exerts immunomodulatory, antiproliferative, and antiviral effects via binding to the type I interferon receptor. The combination is primarily investigated for relapsed low-grade or follicular B-cell non-Hodgkin lymphoma, aiming to enhance anticancer activity above monotherapy responses, with overall response rates (ORR) around 70% and a favorable tolerability profile[1][3][5].
