ZR2 + RDHAP is a sequential chemo-immunotherapy regimen being evaluated for the treatment of newly diagnosed mantle cell lymphoma (MCL). The regimen consists of an induction phase with ZR2 (rituximab, lenalidomide, and zanubrutinib) followed by RDHAP (rituximab, dexamethasone, cytarabine, and cisplatin). Rituximab is a monoclonal antibody targeting CD20; lenalidomide is an immunomodulatory agent targeting the cereblon E3 ubiquitin ligase complex; zanubrutinib is a potent, selective inhibitor of Bruton's tyrosine kinase (BTK). The RDHAP component adds dexamethasone (a corticosteroid), cytarabine (an antimetabolite), and cisplatin (a platinum-based alkylating agent) to provide intensive cytotoxic therapy. This strategy aims to achieve deep molecular remissions in MCL patients.
