Overview

This is a multi-agent combination regimen consisting of four distinct therapies used in the treatment of B-cell lymphomas and related hematologic malignancies. The regimen includes: - **Rituximab**: A monoclonal antibody targeting CD20 on B-cells, leading to their destruction via immune-mediated mechanisms. - **Lenalidomide**: An immunomodulatory agent that enhances immune cell function and has direct antineoplastic effects. - **Zanubrutinib**: A small-molecule, second-generation irreversible inhibitor of Bruton's tyrosine kinase (BTK), blocking B-cell receptor signaling and inhibiting malignant B-cell proliferation[1][5][6]. - **RCHOP** (Rituximab, Cyclophosphamide, Doxorubicin [Hydroxydaunorubicin], Vincristine [Oncovin], Prednisone): A standard chemotherapy regimen for non-Hodgkin lymphoma combining an anti-CD20 antibody with cytotoxic agents and a corticosteroid. This combination is designed to target malignant B-cells through multiple complementary mechanisms—direct cytotoxicity, targeted inhibition of survival pathways, immunomodulation, and immune-mediated cell killing. It is under investigation or use for aggressive or relapsed/refractory forms of non-Hodgkin lymphoma and other high-risk B-cell malignancies.
