Overview

**Rituximab + pegylated interferon α-2b** is a combination regimen investigated for the treatment of advanced indolent B-cell non-Hodgkin lymphoma (NHL) and severe hepatitis C virus (HCV)-related vasculitis. Rituximab, a chimeric monoclonal antibody, targets the CD20 antigen on B-lymphocytes, inducing cell lysis via antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and apoptosis. Pegylated interferon α-2b, a modified cytokine, enhances antiviral and immunomodulatory effects by activating the JAK-STAT pathway, promoting interferon-stimulated gene expression to inhibit viral replication and boost immune responses. Preliminary phase 2 data show the combination is well-tolerated in untreated advanced indolent B-cell NHL, with rituximab biosimilars demonstrating comparable efficacy.
