Overview
R-Pola-Glo is a chemotherapy-light combination regimen being evaluated in the Phase 2 IKF-T062 trial for previously untreated elderly or medically unfit patients with aggressive B-cell lymphomas, including diffuse large B-cell lymphoma (DLBCL). The regimen consists of the anti-CD20 monoclonal antibody rituximab, the CD79b-directed antibody-drug conjugate polatuzumab vedotin, and the CD20xCD3 bispecific antibody glofitamab. Obinutuzumab is also utilized within the protocol, typically as a pre-treatment to mitigate cytokine release syndrome. This combination is designed for patients ineligible for standard R-CHOP therapy, leveraging targeted cytotoxicity and T-cell redirection to treat the malignancy without traditional intensive chemotherapy.
