A combination therapy consisting of rituximab, a chimeric monoclonal antibody targeting CD20 on B cells, and recombinant human thrombopoietin (rhTPO), a thrombopoietin receptor agonist. This combination works through complementary mechanisms - rituximab depletes B cells and reduces autoantibody production that causes platelet destruction, while rhTPO stimulates platelet production by binding to the thrombopoietin receptor and activating the JAK2/STAT5 pathway. The combination has shown superior efficacy compared to rituximab monotherapy in treating immune thrombocytopenia (ITP), with higher complete response rates (45.4% vs 23.7%) and significantly shorter time to response (median 7 days vs 28 days).
