RTM-TDN, also known as rituximab-tetrahedron-doxorubicin conjugate (RTD), is an experimental antibody-DNA nanostructure conjugate (ADNC) being developed by Fudan University Shanghai Cancer Center, Huashan Hospital, and the Eye & ENT Hospital of Fudan University for the treatment of diffuse large B-cell lymphoma (DLBCL). The drug is constructed by conjugating the anti-CD20 monoclonal antibody rituximab to a DNA tetrahedron nanostructure scaffold, which incorporates the chemotherapeutic agent doxorubicin. This design leverages the targeting specificity of rituximab for CD20-positive B cells and the natural affinity of doxorubicin for DNA. Preclinical studies have demonstrated that RTM-TDN exhibits significantly greater anti-tumor potency compared to the standard combination of rituximab and doxorubicin while simultaneously reducing cardiotoxicity and hematotoxicity, offering a potential improvement over the conventional R-CHOP regimen.
