Overview

**RO7490677 + ruxolitinib** is an investigational combination therapy studied primarily for the treatment of myelofibrosis and related myeloproliferative neoplasms. RO7490677, also known as zinpentraxin alfa, is a recombinant human pentraxin-2 protein that targets the bone marrow microenvironment to reduce inflammation and fibrosis. Ruxolitinib is a Janus kinase (JAK) 1/2 inhibitor that acts as an antiproliferative and anti-inflammatory agent by blocking cytokine signaling involved in hematologic malignancies. The combination is intended to synergistically reduce fibrosis and disease burden, with clinical studies indicating a tolerable safety profile and evidence of disease modification. Early trials (Phase II) demonstrate clinical activity and support continued investigation into the use of this combination, particularly for patients with primary myelofibrosis, post-polycythemia vera myelofibrosis, or post-essential thrombocythemia myelofibrosis[1].
