Drug intelligence / Profile preview
RO7836748 is an orally administered small molecule inhibitor developed by Roche that selectively targets mutant RAS G12V proteins, including KRAS G12V, NRAS G12V, and HRAS G12V. The RAS G12V mutation is a well-known oncogenic driver in various advanced solid tumors, such as non-small cell lung cancer (NSCLC), pancreatic ductal adenocarcinoma (PDAC), and colorectal adenocarcinoma (CRC). By selectively binding and inhibiting the mutant RAS G12V proteins, RO7836748 blocks downstream oncogenic signaling pathways to suppress tumor cell proliferation and survival. It is currently being evaluated in a Phase 1 clinical trial (CTR20263852) as a monotherapy and potentially in combination with other anti-tumor therapies for patients with advanced solid tumors harboring the RAS G12V mutation.
