Overview

rQ1-XF is an armed oncolytic herpes simplex virus (oHSV) developed for the treatment of glioblastoma. It is engineered to express two murine cytokines: mXCL1 (lymphotactin) and mFLT3L (Fms-related tyrosine kinase 3 ligand). The mechanism of action involves the selective replication of the virus in tumor cells, leading to oncolysis, while simultaneously secreting mXCL1 to recruit dendritic cells and mFLT3L to promote the expansion and maturation of conventional type I dendritic cells (cDC1s). This dual-cytokine approach is designed to overcome the immunologically cold nature of glioblastoma by enhancing antigen presentation and activating a robust CD8+ T cell-mediated anti-tumor immune response. Preclinical data presented at SNO 2025 demonstrated that rQ1-XF increases cDC1 infiltration and improves survival in specific murine glioblastoma models.
