RTX-IL-12 is an allogeneic red cell therapeutic (RCT) developed by Rubius Therapeutics. It consists of genetically engineered red blood cells that express the cytokine interleukin-12 (IL-12) on their cell surface. By sequestering IL-12 in the vascular system and presenting it in its native form, RTX-IL-12 aims to potently activate and expand T cells and natural killer (NK) cells through cell-cell interactions while minimizing the systemic toxicity typically associated with recombinant IL-12. Preclinical studies have demonstrated its ability to drive Th1 differentiation, increase IFNγ production, and inhibit tumor growth in various models, both as a monotherapy and in combination with checkpoint inhibitors like anti-PD-1.
