**Ruxolitinib + methylprednisolone** is a combination therapy of a small-molecule Janus kinase (JAK) inhibitor (ruxolitinib) and a synthetic glucocorticoid corticosteroid (methylprednisolone), investigated as a first-line or salvage regimen for immune-related diseases, especially **acute graft-versus-host disease (aGVHD)** after allogeneic stem cell transplantation and in relapsed/refractory multiple myeloma. - Ruxolitinib acts as a JAK1/JAK2 inhibitor, blocking pro-inflammatory cytokine signaling, thus modulating T-cell and immune responses. - Methylprednisolone is a potent glucocorticoid, suppressing multiple inflammatory and immune pathways by binding the cytosolic glucocorticoid receptor. The combination demonstrates superior response rates and improved immune regulatory cell recovery compared to steroid-only regimens in aGVHD, and shows well-tolerated efficacy as a salvage option in multiple myeloma.
