**ruxolitinib + NIS793** is an investigational combination therapy comprised of ruxolitinib, a small molecule Janus kinase 1 and Janus kinase 2 inhibitor, and NIS793, a fully human IgG2 monoclonal antibody that binds to and inhibits transforming growth factor-beta 1 and 2 (TGF-β1, TGF-β2), with low affinity for TGF-β3. Ruxolitinib is an approved treatment for myelofibrosis, where it reduces disease symptoms and spleen size by inhibiting dysregulated JAK-STAT signaling. NIS793 modulates the tumor microenvironment by blocking TGF-β signaling, implicated in fibrosis, immune suppression, and the propagation of myeloid cancers. The combination is being developed for patients with myelofibrosis who have a suboptimal response to ruxolitinib alone. This approach aims to improve efficacy by targeting both cellular signaling (JAK-STAT) and fibrotic/immune pathways[1][3][4][7][9].
