ruxolitinib + sacituzumab is a combination therapy consisting of the Janus kinase (JAK) inhibitor ruxolitinib and the TROP2-directed antibody-drug conjugate (ADC) sacituzumab (typically administered as sacituzumab govitecan). This combination is being investigated in the "Umbrella Precision Therapy Study for Bone and Soft Tissue Sarcomas" (ChiCTR2500113769) sponsored by Shanghai Sixth People's Hospital. Ruxolitinib is a small molecule that selectively inhibits JAK1 and JAK2, which are involved in signaling pathways that regulate hematopoiesis and immune function, and is often explored in solid tumors to modulate the inflammatory tumor microenvironment. Sacituzumab govitecan consists of a humanized monoclonal antibody targeting trophoblast cell-surface antigen 2 (TROP2) conjugated to SN-38, a potent topoisomerase I inhibitor, allowing for targeted delivery of chemotherapy to TROP2-expressing sarcoma cells.
