Ruxolitinib + trastuzumab is an investigational combination therapy that was studied for the treatment of metastatic HER2-positive breast cancer. This combination pairs ruxolitinib, a JAK1/JAK2 inhibitor FDA-approved for myelofibrosis, with trastuzumab, an established HER2-targeted monoclonal antibody. The rationale for this combination was based on preclinical evidence showing that the IL6/JAK2/STAT3 signaling pathway is a master regulator in HER2+ breast cancer, and disruption of this pathway reduces HER2+ cell viability. In mouse xenograft models, the combination demonstrated synergistic tumor growth inhibition.
