SAR448501 (also known as DR-0201) is an investigational trispecific natural killer (NK) cell engager developed by Dragonfly Therapeutics and licensed to Sanofi. It is designed using Dragonfly's TriNKET (Tri-specific NK cell Engagement Therapy) platform to treat relapsed or refractory B-cell non-Hodgkin lymphoma (B-NHL). The therapeutic molecule targets CD20 on malignant B cells while simultaneously binding to the NKG2D receptor and CD16 (FcγRIII) on NK cells. This mechanism is intended to activate NK cells and direct them to kill CD20-expressing tumor cells through enhanced antibody-dependent cellular cytotoxicity (ADCC) and direct lysis. SAR448501 is currently being evaluated in a Phase 1 clinical trial for patients with B-NHL who have progressed after at least two prior lines of therapy.
