SB‑1578 is a novel orally bioavailable small molecule inhibitor that targets Janus kinase 2 (JAK2) with specificity within the JAK family and also potently inhibits Fms-like tyrosine kinase 3 (FLT3) and Macrophage colony-stimulating factor receptor (c-Fms) tyrosine kinases. These kinases are involved in signaling pathways central to the pathogenesis of autoimmune diseases such as rheumatoid arthritis. By blocking these kinases and their downstream signaling in relevant cells—including dendritic cells—SB‑1578 inhibits pathological cellular responses associated with inflammation and autoimmunity. Preclinical studies have shown efficacy in rodent models of arthritis by preventing disease onset and treating established disease through reduction of bone resorption and cartilage damage as well as suppression of inflammatory cell infiltration into joints[3][5]. The drug has been investigated in early-phase clinical trials for autoimmune indications[1][2].
