Overview

SBI-087 is a next-generation, humanized Small Modular ImmunoPharmaceutical (SMIP) protein therapeutic directed against the CD20 antigen. It was developed as an anti-inflammatory and antirheumatic agent for autoimmune diseases such as rheumatoid arthritis and systemic lupus erythematosus. SBI-087 works by binding to the CD20 antigen on B cells, leading to their depletion. Preclinical studies showed that SBI-087 resulted in more potent B-cell depletion than rituximab in both peripheral blood and lymphoid tissues. The drug was designed for subcutaneous or intravenous administration and aimed to improve safety, efficacy, and convenience over existing therapies[2][3][5][6][7]. SBI-087 development was discontinued after reaching Phase 2 trials for rheumatoid arthritis and Phase 1 trials for systemic lupus erythematosus due to strategic reasons rather than specific safety concerns[2]. The drug demonstrated effective B-cell depletion but did not progress further in clinical development.
