SCR-A014 is a novel bispecific antibody-drug conjugate (ADC) targeting B7-H3 (CD276) and Delta-like ligand 3 (DLL3), developed by Simcere Zaiming for the treatment of small cell lung cancer (SCLC). The molecule consists of a bispecific antibody conjugated to a topoisomerase 1 inhibitor payload (CPT116). B7-H3 and DLL3 are both highly expressed in SCLC and other neuroendocrine tumors, but they exhibit different internalization kinetics; DLL3 internalizes rapidly while B7-H3 internalizes relatively slowly. By targeting both antigens, SCR-A014 leverages these complementary characteristics to enhance binding capacity and internalization compared to monoclonal ADCs. Preclinical data presented at AACR 2026 demonstrated that SCR-A014 induces potent tumor cell lysis and exhibits significant anti-tumor efficacy in CDX models, suggesting potential as a first-in-class therapeutic strategy for B7-H3 and DLL3 positive cancers.
