sd-rxRNA COX2 is a preclinical RNA interference (RNAi) therapeutic developed by Phio Pharmaceuticals (formerly RXi Pharmaceuticals) using its proprietary self-delivering rxRNA (sd-rxRNA) platform. The compound is designed to silence the PTGS2 gene, which encodes cyclooxygenase-2 (COX-2), a key enzyme in the production of pro-inflammatory prostaglandins. Unlike traditional siRNA, sd-rxRNA molecules are chemically modified to enable spontaneous cellular uptake without the need for delivery vehicles or electroporation. The molecule features a unique asymmetric structure with phosphorothioate modifications and high chemical stabilization. It has been explored for the treatment of proliferative vitreoretinopathy (PVR) via intravitreal administration and as an immunomodulatory agent to reduce immunosuppression in the tumor microenvironment (TME) by targeting myeloid-derived suppressor cells (MDSCs).
