Selinexor + bendamustine + rituximab is an investigational combination therapy composed of: - **Selinexor**: an oral, selective inhibitor of exportin 1 (XPO1), leading to retained nuclear tumor suppressor proteins and apoptosis of malignant cells. - **Bendamustine**: a synthetic alkylating agent with antimetabolite properties, inducing DNA crosslinking and cell death. - **Rituximab**: a monoclonal antibody targeting CD20 on B lymphocytes, mediating antibody-dependent cellular cytotoxicity and complement-mediated cytotoxicity. This combination is under evaluation as a treatment for relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and other B-cell malignancies, showing potential synergistic and additive antitumor effects in preclinical and early clinical studies[2][4][7]. **Main mechanism of action:** antineoplastic, targeted B-cell depletion, DNA crosslinking, and inhibition of nuclear export.
