Overview

This is a combination regimen consisting of **selinexor** (a selective inhibitor of nuclear export, SINE, that targets exportin 1/XPO1 to modulate tumor suppressor protein localization and induce apoptosis), **polatuzumab vedotin** (an antibody-drug conjugate targeting CD79b on B cells, delivering the cytotoxin monomethyl auristatin E for targeted cell death), and **rituximab** (an anti-CD20 monoclonal antibody that induces B-cell lysis by complement-dependent cytotoxicity, antibody-dependent cellular cytotoxicity, and direct apoptosis). This combination is under active investigation for the treatment of relapsed/refractory diffuse large B-cell lymphoma (DLBCL), particularly in patients who have failed prior lines of therapy. The regimen merges targeted cell-killing, immunotherapy, and disruption of nuclear export signaling to maximize antineoplastic effects. The principal developer for selinexor is Karyopharm Therapeutics. Polatuzumab vedotin is developed by Roche/Genentech. Rituximab was originally developed by Genentech/Biogen Idec.
