Overview

A research-stage, tumor-selective **split-genome semi-replicating retroviral gene-therapy system** for glioblastoma. The system comprises two trans-complementing replication-defective retroviral vectors that jointly encode human codon-optimized yeast cytosine deaminase and herpes simplex virus type 1 thymidine kinase. Following intratumoral delivery and propagation in dividing tumor cells, cytosine deaminase converts 5-fluorocytosine into cytotoxic fluoropyrimidine metabolites, while thymidine kinase phosphorylates ganciclovir to its toxic nucleotide form. Dual prodrug administration is intended to generate synergistic tumor-cell killing and bystander effects. Academic and company-associated preclinical studies have evaluated the platform in glioblastoma models, including intracranial xenograft and syngeneic glioma models.
