SGN-40 + rituximab

This drug is a **combination therapy** of two monoclonal antibodies: **SGN-40 (dacetuzumab)** and **rituximab**. SGN-40 is a humanized IgG1 monoclonal antibody targeting CD40, a receptor expressed on B cells; by binding CD40, SGN-40 induces apoptosis and mediates antibody-dependent cellular cytotoxicity (ADCC), primarily through natural killer (NK) cells. Rituximab is a chimeric IgG1 monoclonal antibody directed against CD20 on B cells, mediating B-cell killing via direct apoptosis, ADCC, and complement-dependent cytotoxicity. The combination has been investigated for synergistic or additive antitumor effects in B-cell malignancies such as non-Hodgkin's lymphoma, with the rationale that targeting different B-cell antigens (CD40 and CD20) may produce enhanced tumor cell killing through complementary mechanisms of action[1][3][5].
