Overview

**SHR-A1912 + rituximab** is a combination therapy under clinical investigation primarily for B-cell non-Hodgkin’s lymphoma, specifically diffuse large B-cell lymphoma (DLBCL). SHR-A1912 is a CD79b-targeted antibody-drug conjugate (ADC) consisting of an anti-CD79b monoclonal antibody linked to a DNA topoisomerase I inhibitor, designed to deliver cytotoxic payload to malignant B-cells expressing CD79b[7][5]. Rituximab is a monoclonal antibody targeting CD20, a surface antigen on B-cells, mediating cell death through cytotoxic and immune-mediated mechanisms[4][6]. In clinical trials, SHR-A1912 + rituximab is evaluated with additional chemotherapy agents (e.g., gemcitabine, oxaliplatin), but the combination of SHR-A1912 and rituximab itself constitutes a distinct investigational regimen[3][5].
