Overview

shRNA-CSF1R refers to short hairpin RNA (shRNA) constructs designed to silence the expression of the Colony Stimulating Factor-1 Receptor (CSF1R) through RNA interference (RNAi). In preclinical research, particularly in melanoma, shRNA-CSF1R is used to investigate the functional role of CSF1R signaling in tumor growth, invasiveness, and the acquisition of resistance to BRAF inhibitors. Experimental studies have demonstrated that stable knockdown of CSF1R in melanoma cell lines leads to inhibited cell growth, increased apoptosis, and reduced metastatic potential. This modality is primarily utilized as a research tool for target validation and to provide a preclinical rationale for the use of CSF1R small molecule inhibitors, such as pexidartinib (PLX3397), in clinical settings.
