siG12D-LODER is a miniature, biodegradable polymeric matrix implant containing small interfering RNAs (siRNAs) specifically targeting the KRAS G12D mutation and other G12X mutations in the KRAS oncogene. It is designed for local, sustained release of the RNAi therapeutic directly within solid tumors, particularly pancreatic ductal adenocarcinoma (PDAC). The device is implanted into the tumor via endoscopic ultrasound-guided biopsy needle. By silencing mutant KRAS expression locally, it aims to inhibit tumor growth and enhance antitumor activity when used in combination with standard chemotherapy regimens such as gemcitabine/nab-paclitaxel or modified FOLFIRINOX. Developed by Silenseed, siG12D-LODER has shown promising safety and efficacy signals in early-phase clinical trials for patients with locally advanced or borderline resectable pancreatic cancer[1][3][4][5][6].
