Overview

**siKRAS** is a small interfering RNA specifically designed to silence the expression of the *Kras* gene, including oncogenic and wild-type isoforms, by targeting the 3'-UTR of *Kras* mRNA. By inhibiting KRAS protein synthesis, siKRAS suppresses KRAS-mediated signaling pathways, notably downregulating MAPK activity, inhibiting cell proliferation, and inducing apoptosis in *KRAS*-driven tumors. In preclinical studies using KP mouse models of lung adenocarcinoma, systemic delivery of siKRAS via nanoparticles led to significant reductions in *Kras* mRNA, suppression of downstream phospho-Erk signaling, and tumor regression. siKRAS is typically investigated as a monotherapy or in combination with other small RNAs like miR-34a to enhance anti-tumor efficacy. Its primary indication is the treatment of non-small cell lung cancer and other cancers harboring *KRAS*-mutations[2].
