siKRAS-SWCNT is a preclinical therapeutic candidate developed by Ensysce Biosciences that utilizes single-walled carbon nanotubes (SWCNT) as a delivery vehicle for small interfering RNA (siRNA) targeting the KRAS oncogene. The platform is designed to overcome the delivery challenges of siRNA by complexing it with SWCNTs, often formulated with PEG-lipid analogues to improve circulation half-life and tumor accumulation. In preclinical xenograft models, including MiaPaCa human pancreatic cancer cells, the drug has demonstrated significant target knockdown (up to 90%) and antitumor efficacy. The SWCNT delivery system allows for multi-weekly intravenous administration and leverages the unique near-infrared fluorescence of nanotubes for pharmacokinetic tracking.
