SIL204-SL is an siRNA-based therapeutic candidate developed by Silexion Therapeutics for the treatment of non-resectable locally advanced pancreatic cancer (LAPC) harboring KRAS mutations. It utilizes RNA interference (RNAi) to silence the expression of mutated KRAS, a primary oncogenic driver in pancreatic adenocarcinoma. The therapy is designed for both intratumoral delivery—often via a biodegradable polymeric scaffold (LODER) that provides sustained release—and systemic administration. SIL204-SL is currently being evaluated in an integrated Phase 2/3 trial (PROTACT) in combination with standard chemotherapy regimens, aiming to improve outcomes in patients with KRAS-mutated tumors by inhibiting tumor growth and enhancing sensitivity to chemotherapy.
